SKRINING AWAL EKSTRAK ETIL ASETAT SPONS DYSIDEA SP. SEBAGAI ANTIBAKTERI

Abstract

Penelitian ini bertujuan untuk mengetahui kandungan metabolit sekunder dan kemampuan dari ekstrak etil asetat spons Dysidea sp. dalam menghambat pertumbuhan bakteri Eschericia coli (E.coli) dan Staphylococcus aureus (S.aureus). Isolasi dilakukan dengan ekstraksi menggunakan pelarut metanol:diklorometana (1:1). Campuran metanol:diklorometana disaring, diuapkan dengan rotary evaporator dan kemudian dipartisi dengan etil asetat:air (3:2). Ekstrak kasar etil asetat dianalisis kandungan metabolit sekundernya dengan cara skrining fitokimia. Uji bioaktivitas antibakteri dilakukan dengan metode difusi agar pada konsentrasi 50; 25; 12,5; 6,25 dan 3,125%. Hasil skrining fitokimia menunjukkan adanya senyawa alkaloid dan steroid. Hasil uji antibakteri menunjukkan bahwa ekstrak kasar etil asetat Dysidea sp. mampu menghambat pertumbuhan bakteri E.coli sampai konsentrasi uji 25% dan bakteri S.aureus. sampai konsentrasi uji 3,125%. Hal ini menunjukkan bahwa ekstrak etil asetat spons Dysidea sp. memiliki potensi antibakteri.

Abstract: The purpose of this study was to determine the secondary metabolites and ability of ethyl acetate extract of sponge Leucetta sp. in inhibiting the growth of Eschericia coli (E.coli) dan Staphylococcus aureus (S.aureus). The isolation was performed by extraction using a mixture of methanol:dichloromethane (1:1). Methanol:dichloromethane extracts was filtered, evaporated with a rotary evaporator and than partitioned with ethyl acetate: water (3:2). Crude ethyl acetate extract was analyzed the secondary metabolites by phytochemical screening. Antibacterial testing with agar diffusion method at concentrations of 50; 25; 12,5; 6,25 dan 3,125%. Phytochemical screening results indicate the presence of alkaloid and steroid compounds. The result of antibacterial test showed that the crude ethyl acetate extract of Dysidea sp. able to inhibit the growth of E.coli until the concentration of the test 25% and S.aureus until the concentration of test 3,125%. It means that ethyl acetate extract of sponge Dysidea sp. had antibacterial potency.
